After cooling of the autoclaved vessels containing the nutrient medium, theexplants are inoculated. The actual culture is usually carried out in growth roomsat temperatures of 20–30°C under illumination conditions varying from continuousdarkness to 10,000 lux, from fluorescent lamps. The lids on the vessels are closedby aluminum or paraffin foil, and consequently sufficient air humidity is providedfor at least 4 weeks of culture.
