Typically, FRET-based kinase assays are genetically encoded
fluorescent protein biosensors that can continuously monitor
phosphorylation events in living cells. These biosensors usually
comprise a phosphorylation-site sequence, a protein-binding
domain, and a flexible linker connecting the fluorescent protein
pair. Upon phosphorylation, the protein-binding domain binds to
the phosphorylated residue, brings the FRET pair together into
close proximity and induces a change in FRET ratio.
