We used this system to more precisely define Mre11 requirements and found that deletion of the 26 or 13 C-terminal residues disrupted interaction (Fig. 4b). Furthermore, we asked whether the Mre11 C terminus alone is sufficient for interaction. In this case, the 76 C-terminal amino acids of Mre11 (Mre11c76aa) were expressed in the yeast two-hybrid system, along with CDK2. We found that the final 76 amino acids of Mre11 can interact with CDK2, although with lower apparent affinity than full-length Mre11 (Fig. 4c).
